Study of Loss of Heterozygosity on the Loci THRB of Chromosome 3 in Patients with Renal Cell Carcinoma

Abstract
Using a polymerase chain reaction(PCR) based restriction fragment length polymorphism analysis to histopathologicall proven 35 samples of matched blood and tumor tissue with sporadic RCC, LOH at THRB locus on the short arm of chromosome 3 which
has
multiple recurrence genes was investigated. Allelic analysis was compared with the histopathological findings to correlate the relationship between oncogenesis and genetic alterations.
1. In the allelic frequency of three loci, THRB(EcoRI), THRB(BamHI) and THRB(MspI), the heterozygosity was seen in 19 (54.3%) out of 35 cases on THRB(EcoRI) locus. Of 19 heterozygosity, LOH was seen in 2 cases(10.5%). The heterozygosity was seen
in 16
(59.3%) out of 27 cases on THRB(MspI) locus. Of 16 heterozygosity, LOH was seen in 1(7.1%). However, the heterozygosity was seen in only 2 (5.7%) out of 35 cases on THRB(BamHI) locus and LOH was not seen in THRB(BamHl).
2. Showing the rate of LOH according to the cell type, the cell type, the clear distinction was shown between clear and granular cell type. LOH was not observed in clear cell type whereas in granular cell type. LOH was observed in near telomere
about
33.3% each other in THRB(EcoRI) and THRB(MspI) locus but could not be observed in THRB(BamHI) locus.
This study showed the possiblility of multistep tumorigenesis by revealing high incidence of LOH other than existing loci, Also, there was weak correlation between genetic analysis histopathological findings.
